Polarizing macrophages derived from human THP-1 cells in vitro: methods and protocols
Abstract: Macrophages derived from the human THP-1 cell line have been widely used as substitutes for primary macrophages in various macrophage-related studies. However, difficulties still exist in establishing THP-1 macrophage models. This research presents techniques for generating different phenotypes of activated macrophages derived from THP-1 cells by introducing specific stimuli and provides some potential markers to confirm each type of activated macrophage. It is hoped to provide novel and useful methods for scientific research and to help researchers explore this field more intuitively and effectively.

摘要:巨噬细胞具有高度的可塑性与异质性,能在特定微环境中调控细胞增殖、介导免疫调节并促进组织再生。在相关研究中,由人源THP-1细胞系诱导分化而来的巨噬细胞,已被广泛用作原代巨噬细胞的替代模型。然而,在诱导建立THP-1巨噬细胞的各类模型时,仍面临诸多挑战。本研究详细阐述了通过特定刺激将THP-1细胞诱导为不同表型巨噬细胞的方法,并筛选出一系列可用于识别不同巨噬细胞表型的潜在标志物,旨在为该领域研究提供新颖且实用的技术手段,助力研究人员更直观、高效地开展探索。
