创新点:探讨了老年(50~80岁)膝骨关节炎患者白细胞mtDNACN和血浆炎性细胞因子水平及二者的关系.
方法:分别收集膝骨关节炎组和对照组血液样本并对膝关节评分(Kellgren-Lawren grading).使用实时定量聚合酶链反应(qRT-PCR)检测相对mtDNACN;使用多重免疫分析(multiplex immunoassay)测定血浆中10种炎性细胞因子水平;应用线性相关、Logistic回归和主成分分析(PCA)揭示骨关节炎白细胞mtDNACN和血浆炎性细胞因子的相关性.
结论:血浆中白介素4(IL-4)、IL-6和全血白细胞mtDNACN可能是膝骨关节炎有效的生物标志物;IL-5则对mtDNACN减少具有潜在的影响.

创新点:定量研究了草莓对隔离环境中人的生理、心理及认知的影响,并在研究中同时纳入了实验室短期模拟隔离实验和真实环境下的长期隔离实验.
方法:在实验室实验中,通过测量心率、唾液皮质醇和心理量表等指标,并与草莓模型和静坐对比,研究观察草莓对情绪的影响;采用注意广度、短时记忆广度和选择反应时测试,进一步探究草莓对认知的影响.基于实验室实验的结果,在生物再生生命保障系统中进行长期隔离实验,进一步验证隔离环境中草莓对人情绪的正向调节作用.
结论:本研究中,在实验室短期模拟隔离实验和真实环境下的长期隔离实验下,观察草莓15分钟可降低皮质醇水平,并减轻紧张和困惑等负面情绪.同时,15分钟的草莓观赏使隔离环境中人的选择反应时显著降低,反应力明显加快,有助于警觉性的提高.

方法:应用流式细胞术检测CFZ对肺腺癌细胞周期和 凋亡的影响;通过MTS比色法及平板克隆形成实验分析CFZ对肺腺癌细胞生长的抑制作用;使用蛋白质印迹法(western blot)和定量聚合酶链反应(qPCR)检测相关基因表达水平的改变.
结论:CFZ通过AKT/FOXO3a通路上调Gadd45a基因的表达,诱导肺腺癌细胞周期阻滞和凋亡,从而发挥抗肿瘤效应.

创新点:首次报道了3-BrPA对秀丽隐杆线虫有毒性作用,己糖激酶是3-BrPA对线虫作用的重要靶点,而CYP-35A家族是线虫代谢3-BrPA的主要酶类.
方法:用不同浓度的3-BrPA处理秀丽隐杆线虫,每24 h监测一次存活率;用实时荧光定量聚合酶链反应(qPCR)检测代谢相关基因的表达;通过RNAi沉默己糖激酶家族基因hxk-1、hxk-2和hxk-3;计算3-BrPA处理hxk家族RNAi株和细胞色素P450 cyp-35家族突变株后的致死中浓度(LC50).
结论:3-BrPA对线虫有明显的毒性效应(图1);与对照组比较,3-BrPA处理组的线虫平均寿命明显缩短(图2);3-BrPA处理线虫后hxk-1、hxk-2和hxk-3的信使RNA(mRNA)表达明显升高(P<0.05,图5);3-BrPA处理hxk RNAi株后的LC50均减小(P<0.05,表5);3-BrPA处理cyp-35突变株后的LC50也均减小(P<0.05,表6).综上所述,3-BrPA的毒性与其对秀丽隐杆线虫己糖激酶代谢的影响密切相关;CYP-35家族在线虫中对3-BrPA代谢中起着关键作用.

创新点:既往有隐睾下降固定术手术史的非梗阻性无精子症的患者睾丸局部粘连严重,所以睾丸显微取精术时睾丸暴露困难,因此手术难度明显增大.术者将常规睾丸显微取精术改良为分步法小切口睾丸显微取精术,术中无需完全游离暴露睾丸,仅通过小切口睾丸显微取精术就可使一半的患者成功获取精子.因此分步法小切口改良睾丸显微取精术可在不降低睾丸取精成功率的前提下缩短手术时间和减少手术创伤.
方法:收集2015年5月至2017年3月于上海交通大学附属第一人民医院泌尿中心男科接受睾丸显微取精术的20例既往有隐睾下降固定术手术史的非梗阻性无精子症,采用分步法小切口改良睾丸显微取精术获取睾丸精子.分步法小切口改良睾丸显微取精术具体步骤为:第一步,小切口睾丸取精术;第二步,将小切口延长为标准切口,行标准睾丸显微取精术;第三步,对侧睾丸标准睾丸显微取精术.
结论:小切口睾丸显微取精术可快速有效获取睾丸内表浅位置饱满的生精小管,标准的睾丸显微取精术可作为小切口睾丸显微取精术失败时的补救措施.分步法小切口睾丸显微取精术可降低睾丸损伤前提下,快速有效地获取睾丸精子.这种手术方式特别适用于睾丸内存在表浅饱满生精小管的非梗阻性无精子症,如隐睾下降固定术后的非梗阻性无精子症.
