Abstract: 

Intracellular 3D printing

目前细胞工程研究正从“让细胞表达产物”转向“直接制造细胞内的功能结构”。本研究提出基于双光子成形的生物相容材料的悬浮细胞内直接打印方法,以实现活细胞内可编程的微米级三维功能结构的制造,从而克服简单胞吞或吞噬作用难以吞噬微米级大颗粒的局限。本项研究重点介绍了将生物相容性光引发剂大批量渗透进细胞与自动化双光子打印技术相结合的工作流程,并讨论如何将内源性蛋白、生物相容性单体或其他生物材料作为交联元件纳入上述体系中,以减轻免疫排斥和细胞毒性。这项研究将细胞从单纯的反应容器提升为可以进行操控的微型工厂,进而直接影响细胞内传感、追踪和精准药物递送。目前这项新技术依然面临诸多关键挑战,包括建立标准化可用于活细胞内打印的材料库、实现自动对焦和细胞姿态的自适应控制,以及根据细胞功能来协同设计打印装备等。预计“细胞内3D打印”技术将为合成生物学与微纳生物制造之间提供一种新颖的接口。
